Expression of an α-expansin involved in guava (Psidium guajava L.) fruit ripening

Abstract

The partial segment of a cDNA, designated PgEXP1, coding for a homologue of expansins, was isolated and characterized from ripe guava fruit (Psidium guajava L.). The bioinformatics analysis revealed that PgEXP1 contains part of two domains characteristic of the α-expansins. Also, the phylogenetic tree shows a great relationship with the α-expansin of tomato (Solanum lycopersicum L.) which is specific for fruit ripening. Expression studies by RT-PCR performed in different tissues of the plant and fruit ripening of eight states, showed that the expression is only in ripe fruit. Expression in eight different states of ripeness was analyzed by Northern blot and found mainly in the last three states. The analysis of these results shows that the PgEXP1 is specific for guava fruit ripening.
